Abstract
This investigation demonstrates that electrophoretically cryptic genetic variation involving methionine residues, can be resolved using site-specific cleavage of the polypeptide chain with cyanogen bromide, followed by SDS-polyacrylamide electrophoresis of the fragments. Orosomucoid, also called α₁-acid glycoprotein, was used as the model to test this approach because the proportion of methionine residues has been reported to be less than unity in this protein, evidence that can be interpreted as due to heterogeneity. A method to purify the model protein, orosomucoid, from as little as 0.5 ml of serum was developed,as a first step in this investigation. Genetic analysis of familial and population material confirmed a polymorphism, Val/Met, at residue 156.
